TGFB-1 (Transforming Growth Factor Beta 1) and TGFB-2 (Transforming Growth Factor Beta 2) are members of the TGFB superfamily of cytokines, which play critical roles in cellular processes including proliferation, differentiation, and migration, as well as immune responses. Both TGFB-1 and TGFB-2 are synthesized as precursor proteins that are proteolytically processed to release the mature, active cytokine. They signal through a heteromeric complex of serine/threonine kinase receptors, leading to the phosphorylation and activation of SMAD transcription factors, which in turn regulate the expression of target genes. TGFB-1 is predominantly involved in immune regulation, wound healing, and fibrosis. It has potent immunosuppressive properties and is critical for the maintenance of immune homeostasis. TGFB-2, while sharing many functions with TGFB-1, has distinct roles in the development of the nervous system and the regulation of cellular differentiation in various tissues. Both cytokines are involved in pathophysiological conditions, such as cancer, where they can promote tumor growth and metastasis by modulating the tumor microenvironment.
